Folinic acid protection against nitrous oxide teratogenicity in the rat.

چکیده
The Sprague-Dawley rat was used to demonstrate the effect of nitrous oxide, with and without folinic pretreatment, on reproductive indices and fetal development. One of the objectives of the investigation was to test the hypothesis that at least some of the teratogenic effect of nitrous oxide is related to interference with folate metabolism. Groups of animals were exposed to 70-75% nitrous oxide on day 9 of pregnancy with or without folinic acid 0.1 mg i.p. 12 h before, and immediately before, exposure. Subsequent fetal development was compared with that of various control groups. There were no significant differences in fetal survival, but fetal weights were reduced in both groups exposed to nitrous oxide. Of the indices of skeletal maturity, the number of ossified sternebrae was reduced only in the nitrous oxide group not receiving folinic acid. The incidence of major skeletal abnormalities in the untreated nitrous oxide group was significantly increased to five times that of the control groups, whereas the incidence in the nitrous oxide group receiving folinic acid was not significantly different from control. It is concluded that pretreatment with folinic acid can at least partially reduce the teratogenic effects of nitrous oxide in the rat.

Prevention of nitrous oxide-induced megaloblastic changes in bone marrow using folinic acid.
Prolonged anaesthesia with nitrous oxide inactivates vitamin B12 and impairs DNA synthesis in bone marrow cells. The use of parenteral folinic acid in the prevention of these toxic effects has been studied in 11 patients, ventilated artificially with nitrous oxide in oxygen for 24 h. Bone marrow aspirates were performed before and after exposure to nitrous oxide. They were assessed morphologica...
متن کاملMegaloblastic bone marrow changes after repeated nitrous oxide anaesthesia. Reversal with folinic acid.
A seriously ill patient was found to have megaloblastic changes in his bone marrow after a nitrous oxide anaesthesia which lasted for 105 min. After an interval of 7 h a second nitrous oxide anaesthetic was administered, during which the patient received 30 mg of folinic acid. His marrow was normal when assessed 4 h later.

متن کاملNitrous oxide (laughing gas) facilitates excitability in rat hippocampal slices through gamma-aminobutyric acid A receptor-mediated disinhibition.
ALTHOUGH nitrous oxide is safe when administered by anesthesiologists, misuse may result in brain damage either via asphyxia or via direct effects resulting in vacuole formation in cingulate and retrosplenial cortices under normoxic conditions. The direct toxic effects of nitrous oxide raise the possibility that nitrous oxide toxicity results from excitatory effects of the drug. To determine ho...

Background: Methanol poisoning may result to death and permanent complications, sporadic cases of methanol poisoning are fairly rare and the largest experience are generally gained from managing methanol intoxication epidemics. The main metabolite of methanol is acid formic that is the prime metabolite responsible for toxic effects of methanol and since folate dependent systems are responsible ...
